分子生物学,生物化学,细胞生物学,遗传学及表观遗传学,非编码RNA (noncoding RNA),基因表达及调控,癌症生物学。

研究经验和研究领域:

1)药用植物抑癌活性成分的药效机理研究:药用植物中的很多成分具有抑癌活性,但其药效机理不明,限制其临床应用。为突破该瓶颈,我们将探究植物药用成分在肝癌细胞内的分子靶点、所影响的重要基因及细胞生物途径,创立科学的联合给药方式以有效抑制肝癌发展并降低对机体副作用。首次在细胞水平验证了鞣花酸联合阿霉素或联合顺铂的抗肝癌活性,并在小鼠模型中鉴定了鞣花酸联合阿霉素使用,可以显著加强阿霉素抗肝癌活性,减弱由高浓度下阿霉素引发的心肌毒性。此外,我们还研究了百里香琨,地榆皂苷II的植物天然活性物质的抑癌活性。

2)发明了以DNA载体为基础的RNA干扰(RNAi)技术(即shRNA方法)。这一技术的发明为许多生物学科的研究提供了全新的方法,尤其是对基因和蛋白质功能的研究具有重大的创新性,也进了RNAi技术在医疗上的应用。目前,这一方法已经成为世界范围实验室用于研究基因功能的常规实验技术。该方法的DNA载体成为Ambion/Life Technologies公司的第一代产品(商品名pSilencer)。做为第一作者的相关PNAS文章已被引用上千次。

3)利用对Yin Yang 1(YY1)蛋白在肿瘤发生中功能的研究来探索几种常见癌症(乳腺癌,前列腺癌,肺癌)的致病机制。我的多年的研究成果确立了YY1的癌基因地位。作为一个多功能蛋白,YY1在癌症发生中的终极作用一直有争议。我在2004年发表的Cell文章中发现了YY1对p53的负调控,奠定了YY1的癌基因的地位。这些研究进一步确定YY1的癌基因地位,并揭示了YY1在癌症治疗中作为靶基因的可能性。通过干扰YY1与癌蛋白之间相互作用以抑制肿瘤生长的多肽已被授权专利。

4)对其它抑癌基因、原癌基因及非编码RNA的研究:(1)利用基因芯片等方法对抑癌基因SOX7的研究,发现其上、下游调控因子及可能的调节机制;(2)对DMTF1研究,发现了这一基因产生mRNA的不同选择性剪接(alternative splicing)形态对乳腺癌发生和发展具有完全相反的作用,以及调节该可变剪接的分子机制;(3)研究了几种microRNA对原癌基因EZH2、YY1和FASN的调节作用;(4)利用多个乳腺癌及它们对应的癌旁组织样品中的长非编码RNA(lncRNA)的分析,发现了多种在乳腺癌中差异性表达的lncRNA,目前正在对它们的生物学意义和对乳腺癌的调节功能进行鉴定。
